Material and Methods Materials. Neurobasal medium, B27, SUPERSCRIPT II RNase H reverse transcriptase, RNAaseOUT, and Taq polymerase were from Invitrogen (Paisley, Scotland); papain was from Worthington Biochemical Corp. (Lakewood, NJ); NaAsO2, cysteine, soybean trypsin inhibitor (STI), propidium iodide (PI), Hoechst 33342, and the bicinchoninic acid protein assay kit were from Sigma (Poole, Dorset, UK). All other reagents were from Invitrogen or Sigma. The following antibodies were used: anti-ERK1/2 (clone MK12; 1:5000); anti-KDEL (1:500) from Abcam (Cambridge, UK); anti-Puma (3043; 1:1000) from Prosci (Poway, USA); antiBiP/grp78 (1:1000) from BD transduction labs (New Jersey, USA). Anti-total p73 (1:2000) was a gift from Dr Gerry Melino (MRC Toxicology Unit, Leicester, UK); anti-CHOP/Gadd153 (1:250) was a gift from Dr Linda Hendershot (St. Judes Childrens Research Hospital, Memphis, USA). All anti-mouse and anti-rabbit horseradish peroxidase-conjugated were from Jackson ImmunoResearch Laboratories (West Grove, PA).
